Isaac was the son of John Prillaman (1754-1842) and his first wife (name unknown).
Isaac's father John was the son of Jacob Prillaman, Sr. (1721-1796) and his wife Waltpurgelly. Jacob was the first Prillaman in Virginia, arriving into Philadelphia in 1747 from his home in Switzerland, migrating to Maryland, and eventually settling in Franklin County, Virginia. Jacob was a Revolutionary War Patriot.
Isaac married his first cousin, Ruth Prillaman, daughter of Daniel Prillaman and Ann O'Bryant. They were married 5 Jan. 1824 (Bond) in Franklin County, Virginia., with surety provided by Thomas Helms. The approximate value of Isaac's property in 1860 was twenty seven thousand dollars. Isaac and Ruth had one child.
Isaac was bequested in his father John Prillaman's will as follows:
"To my son Isaac I give all my land on the South side of the Long hollow Ridge, the Summit of the ridge to be the dividing line between him and the land herein after devised to my son Abraham and my daughter Susannah, to my said son Isaac and his heirs forever."

This Prillaman Cemetery is located about 2 miles from Callaway on the property of Robert Prillaman, it is a different cemetery from the other original Prillaman cemetery at Callaway
