Jacob Prillaman was the son of John Prillaman and his first wife (name unknown) of Franklin County, Virginia.

John was the son of Jacob Prillaman, Sr. (1721-1796) and Waltpurgelly. Jacob, Sr. was the first Prillaman in Virginia and a Revolutionary War Patriot.

Jacob received the following bequest from his father John's will dated 1838 and probated in 1842 in Franklin County, Virginia as follows:

"to my son Jacob I give the sum of Sixty Seven Dollars, to be paid to him in money by my son Abraham which will make with what he has heretofore received , the sum of about five hundred dollars and no more."
When Jacob's father John's will was probated in 1842, Jacob had been away from his Franklin County home for about seventeen years.

"Jacob left Virginia , and settled in Rhea County, Tennessee circa 1825, where he appeared on the 1830 census. Jacob had married his first wife, Elizabeth Grayson, about 1806, in Virginia. Elizabeth was the daughter of William and Rachel (Cooley) Grayson of Montgomery County, Virginia. Jacob and Elizabeth had ten children, eight or nine of the children were born in Virginia, and the tenth child was born in 1829 in Rhea County, Tennessee. Elizabeth died in 1830 in Rhea County.

Jacob married his second wife Desdamony Gorman on 23 September 1833 while still living in Rhea County, Tennessee. They had two children, the first child was born on a flatboat on the Tennessee River while Jacob and Desdamony were in route to Arkansas in the summer of 1835 (name of this child was Eliza Jane). In the fall of 1835 Jacob and family settled in Hot Springs County, Arkansas. Their second child (Jacob Prillaman, Jr.) was born in 1839. Jacob's second wife Desdamony died five years later in 1840.

In 1842, Jacob and family left Hot Springs County, and moved to Saline County, Arkansas.
In Saline County, he married his third wife Sarah Brazil on 29 October 1842. Sarah was born in 1799 in Kentucky. She had two children by her previous marriage. Jacob and Sarah did not have children. Jacob Prillaman owned about a dozen slaves when he moved to Saline County. He lived in Saline County for about ten years. He died on 20 November 1852, at about age seventy.
His estate was divided equally among his children in Arkansas, but died intestate. His widow and third wife Sarah A. Brazil Prillaman remarrried a Mr. Giles. She died 18 November 1855, three years after Jacob died.
Jacob was most likely interred on his farm in Saline County, where there is a mountain called "Prillaman Mountain", from the base flows a cold mountain spring. Jacob had a total of twelve children, ten with his first wife Elizabeth, and two with his second wife Desdamony.
