Daniel Prillaman married Ann O'Briant about 1777. Both were 19, Ann being the elder by 10 months. They likely grew up together and would produce a family of ten. On 17 June 1783, Daniel and Ann received a grant of 285 acres on Nicholas Creek adjoining Ann's father's land. Eventually Dennis O'Briant would by deed of gift transfer his land of 286 acres to his "son Daniel Prillaman." In turn, the O'Briant tract of land was passed on to David Prillaman, son of Daniel and Ann. On this land, still in the family today, rest many of the Prillaman and O'Briant descendants.

Daniel was industrious, first running a powder mill which exploded, possibly causing him to turn to more mundane occupations and appointments such as supervisor of county road surveys, overseer of the poor, and sitting as a juror.

Daniel's will was probated in 1854 and names his wife and ten children. Witnesses to Daniel's will were G. B. Lesueur, James W. Lesueur and Thomas H. Prillaman.
